Het Satsuma dwergvirus (SDV) beschadigt de citrusproductie ernstig door de kwaliteit en opbrengst van fruit te verminderen. Om besmetting met SDV te voorkomen, worden moederbomen voorafgaand aan de distributie van boomkwekerijen gecontroleerd op SDV-vrij. In deze studie hebben we een immunochromatografische assay (ICA) -kit vergeleken met een dubbel-antilichaam sandwich enzymgekoppelde immunosorbent-assay (DASELISA) voor grootschalige diagnose van SDV in boomgaardbomen in de prefectuur Shizuoka, Japan.
De twee methoden gaven tegenstrijdige resultaten voor 11 van de 1.705 monsters, die allemaal negatief waren volgens DAS-ELISA maar positief waren door ICA. De monsters die door DASELISA of ICA als positief werden gescoord, werden geanalyseerd door middel van reverse-transcriptie-polymerasekettingreactie en ze werden allemaal als positief bevestigd. Deze resultaten bevestigen het gebruik van ICA als screeningsmethode voor grootschalige diagnose. Stamdiscriminatie onthulde dat 16 van de 22 isolaten tot SDV behoorden, terwijl alleen infectie met citrusmozaïekvirus (CiMV) en co-infectie (SDV en CiMV) in de minderheid waren.
Deze studie had tot doel te onderzoeken of de trypsine-achtige peptidase-activiteitsassaykit (TLP-AA-Kit), die de activiteit van N-benzoyl-dl-arginine peptidase (trypsine-achtige peptidase) meet, kan worden gebruikt als een betrouwbaar hulpmiddel voor parodontitis detectie in populatie-gebaseerde surveillance. In totaal ondergingen 105 personen een parodontaal onderzoek over de hele mond en verstrekten ze tonguitstrijkjes als specimens voor verdere analyses. De resultaten van de TLP-AA-Kit werden gescoord tussen 1 en 5; hogere scores duidden op hogere trypsineconcentraties. Analyses van de werkingskenmerken van de ontvanger werden gebruikt om de voorspellende validiteit van de TLP-AA-Kit te evalueren, waarbij de parodontitisgevaldefinitie van de Centers for Disease Control / American Academy of Periodontology als referentie diende.
Ernstige en matige parodontitis werden vastgesteld bij respectievelijk 4,8% en 16,2% van de onderzoekspopulatie. De TLP-AA-Kit toonde een hoge diagnostische nauwkeurigheid voor ernstige parodontitis, met een oppervlakte onder de curve van 0,93 (95% betrouwbaarheidsinterval = 0,88-0,99). De diagnostische nauwkeurigheid van de TLP-AA-Kit voor matige / ernstige parodontitis was echter niet betrouwbaar. Hoewel verdere studies nodig zijn om onze resultaten te valideren, benadrukken de hierin verstrekte resultaten het potentieel van de TLP-AA-Kit als een nuttig hulpmiddel voor de detectie van parodontitis, vooral in ernstige gevallen, voor populatie-gebaseerde surveillance.
Vergelijkende prestaties van SARS-CoV-2-detectietesten met zeven verschillende primer- / probesets en één assaykit
More than 100,000 people worldwide are known to have been infected with SARS-CoV-2 beginning in December 2019. The virus has now spread to over 93 countries including the United States, with the largest cluster of US cases to date in the Seattle metropolitan area in Washington. Given the rapid increase in the number of local cases, the availability of accurate, high-throughput SARS-CoV-2 testing is vital to efforts to manage the current public health crisis. In the course of optimizing SARS-CoV-2 testing performed by the University of Washington Clinical Virology Lab (UW Virology Lab), we tested assays using seven different primer/probe sets and one assay kit.
We found that the most sensitive assays were those the used the E-gene primer/probe set described by Corman et al. All assays tested were found to be highly specific for SARS-CoV-2, with no cross-reactivity with other respiratory viruses observed in our analyses regardless of the primer/probe set or kit used. These results will provide invaluable information to other clinical laboratories who are actively developing SARS-CoV-2 testing protocols at a time when increased testing capacity is urgently needed worldwide.
Vergelijking van commerciële ELISA-kits, een prototype multiplex elektrochemoluminescentietest en een multiplex op parels gebaseerde immunoassay voor het detecteren van een op urine gebaseerde, blaaskanker-geassocieerde diagnostische handtekening.
De mogelijkheid om nauwkeurig meerdere eiwitten tegelijk te meten in een enkele test, kan de efficiëntie van klinische tests die uit meerdere biomarkers bestaan aanzienlijk verbeteren. We onderzochten de diagnostische nauwkeurigheid van de twee multiplex eiwitarray-platforms voor het detecteren van een met blaaskanker geassocieerde diagnostische signatuur in monsters van een cohort van 80 proefpersonen (40 met blaaskanker). Urinemonsters uit de banken verzameld van de universiteiten van Kyoto en Nara werden vergeleken met histologisch bepaalde blaaskanker.
De concentraties van de 10 eiwitten (A1AT; apolipoproteïne E-APOE; angiogenine-ANG; koolzuuranhydrase 9-CA9; interleukine 8-IL-8; matrixmetalloproteïnase 9-MMP-9; matrixmetalloproteïnase 10-MMP10; plasminogeenactivatorremmer 1- PAI-1; syndecan-SDC1; en vasculaire endotheliale groeifactor-VEGF) werden gecontroleerd met behulp van twee prototype multiplex array-platforms en een enzymgekoppelde immunosorbenttest (ELISA) volgens de technische specificaties van de fabrikant. Het bereik voor het detecteren van elke biomarker werd verbeterd in de multiplexassays, hoewel de ondergrens van kwantificering (LLOQ) doorgaans lager was in de commerciële ELISA-kits.
Het gebied onder de werkingskenmerken van de ontvanger (AUROC) van de prototype multiplex-assays was naar verluidt 0,97 voor de multiplex parel-gebaseerde immunoassay (MBA) en 0,86 voor de multiplex elektrochemoluminescente assay (MEA).
